Abstract 【Abstract】 Objective To explore the correlation between transforming growth factor-β(TGF-β)/
leukemia inhibitory factor (LIF) signaling pathway and the regulation of cyst formation of hepatic
echinococcus granulosus in children. Method From January 2020 to June 2021, 40 children with hepatic echinococcosis were selected from the Department of General Surgery, Children's Hospital of Xinjiang Uygur Autonomous Region. Cyst removal surgeries were performed, and the protoscolices were isolated and cultured in vitro. There were three study groups which were added with LIF factor, LIF siRNA and Smad4 siRNA respectively, and one control group. After two weeks of cultivation, the protoscolices withcystic characteristics were examined under the microscope, and total protein was extracted. Enzyme-linked immunosorbent assay was used to test the expression levels of TGF-β, Smad2, Smad3, Smad4 and LIF.One-way analysis of variance and t-test were used as statistical methods. Result There were statistically significant differences in the expression levels of TGF-β, Smad2, Smad3, Smad4, and LIF among the groups (all P<0.001). The expression levels of the above five indicators in the control group were (3 152±276) ng/L and(17.4±3.0), (15.9±0.9), (18.6±1.8), and (1 334±236) μg/L. The expression levels of TGF-β, Smad2,Smad4, and LIF [(4 100±173) ng/L and (34.3±3.0), (26.3±2.2), and (1 984±237)] μg/L] in the LIF group were all higher than those in the control group (all P<0.05). The expression levels of the above five indicators in the LIF siRNA group [(2 151±136) ng/L and (9.2±1.2), (12.2±0.8), (12.8±0.4),and (464±244)] μg/L] were lower than those in the control group (all P<0.05). The expression levelsof the above five indicators in the Smad4 siRNA group [(2 529±267) ng/L and (5.7±1.3), (10.1±0.7),
(9.8±2.0), and (246±53)] μg/L] was also lower than those in the control group (all P<0.05). Conclusion Invitro experiments have shown that TGF-β/LIF signaling pathway may be related to the regulation of cyst formation of hepatic echinococcus granulosus in children. The development of protoscolices towards cyst formation may be regulated by LIF.
