Abstract 【Abstract】0bjectives To explore the effect of miR一20b over—expression on the proliferation of P19
cells and the differentiation of P19 cells into myocardiocyte.Methods P19 cells were infected with the lenti—viral vector containing nlil{lU—miR~20b and the empty vector.respectively.Stably miR一20b over—ex—pressed P19 cell line was successfully constructed by puromycin screening for two weeks.which wasvalidatedby fluorescent observation and real~time quantitative PCR.The proliferation of P19 cells was analyzed bvCCK一8 method.Two cell groups were cultured in FBS—free—Or.一MEM for 24h to make them both at Ca)phase,which were assayed cell cycle by flow cytometry.Then we detected the relative expression of soll{1e heartrelated marker genes by real—time quantitative PCR and observed the cell morphology under light microscopeafter inducing P19 cells into cardiomyocytes under 1%DMSO.Results We Successfullv established stablvinfected P19 cell line and miR一20b was significantly over—expressed.Compared with the empty vector.miR一20b over—expressed could suppress the proliferation of P19 cells and promote the relative exDression of related marker genes(P<0.05,P<0.010rP<0.001).Conclusion MiR一20b could inhibit the Dr0一lireration of P19 cells and promote the cardiomyocytes difierentiation.
