The dynamically changes of Uncoupling protein 2 in myocardial tissue of septic rats |

Abstract 【Abstract】Objective To investigate the relationship between uncoupling protein 2(UCP2)expression
and the septic myocardial injury-.Methods Forty SD male rats were randomly divided into 5 groups:control group,6 hour—sepsis group,12 hour—sepsis group,24 hour—sepsis group and 48 hour—sepsis group.The sepsis model was established by intraperitoneal injection of lipopolysaccharide 10 rag/kg.Each group were sac—rificed at the appropriate timepoint.The expression of UCP2 in myocardium was measured by Western Blot—ring.RT—PCR and hnmunohistochemistry-.Results After modeling,UCP2mRNA gradually increased be—ginning at 6 hour—sepsis group,at 24 hour—sepsis group up to the peak value.Sepsis 6,12,24,48 hgroupmyocardial UCP2mRNA was 1.31±0.06,2.94±0.40,8.20±1.96 and 2.15±0.24.The 12,24,48hseptie groups increased significantly compared with that in control group[(0.78±0.22)P<0.05 or P<0.01].The changes in UCP2 protein levels are similar to mRNA levels.Sepsis 6h,12h,24h,48hgroupsUCP2 protein level was(0.0806±0.0067,0.1063±0.0054,0.1200±0.0057 and 0.0985±0.0096)were significantly higher than those in control group,0.0516±0.0080(P<0.01).Conclusion septic rat myocardial tissue uncoupling protein 2 up—regulated,this change may be involved in the pathophysiology-of myocardial injury-in sepsis process that requires further study its mechanism.
