【Abstract】 Objective To explore the clinical significance of microRNA-34a(miR-34a) expression inplacenta of the patients with preeclampsia. Methods Thirty patients with preeclampsia in Dongguan People'sHospital between September 2017 and June 2018 were enrolled in this study. Another 30 cases of normalcesarean section pregnant women were selected as control group. Real-time PCR was used to detect miR-34aand myeloid cell leukemia-1(MCL-1). MCL-1 protein and microtubule associated protein 1 lightchain 3 Ⅱ(LC3 Ⅱ ) were detected by Western blot. Quantitative data were compared with t test, and the comparativestudy was carried by Spearman correlation analysis. Results In preeclampsia group,the 2- △ △ Ct valueof miR-34a was 0.217±0.035, which was significantly higher than that of 0.126±0.028 in control group(P<0.01). The 2- △ △ Ct value of MCL-1 in preeclampsia group was 0.208±0.037, which was significantlylower than that of 0.316±0.054 in control group (P<0.01). The expression of miR-34a in preeclampsiagroup was negatively correlated with MCL-1 expression (r=-0.412, P<0.05). The gray level of MCL-1protein in preeclampsia group was 0.413±0.072, which was significantly lower than that of 0.826±0.142 incontrol group (P<0.01). The gray level of LC3 Ⅱ protein in preeclampsia group was 0.316±0.048, whichwas significantly higher than that of 0.152±0.024 in control group (P<0.01). Spearman correlation analysisshowed that there was a significant negative correlation between LC3 Ⅱ protein expression and MCL-1 protein expression (r=-0.607, P<0.05), and LC3 Ⅱ had a significant positive correlation with miR-34a
expression (r= 0.582, P<0.05). Conclusion High expression of miR-34a in placenta of preeclampsia may
inhibit MCL-1 gene to promote autophagy, and participate in the pathogenesis of preeclampsia.
