reticulum stress,ERS)的关系,以及其在子痫前期致病中的作用。 方法 2018 年10 月至2019 年9 月
在海南医学院第二附属医院产科收治的子痫前期患者30 例为研究对象,同时选择同期在本院正常分
娩产妇30 例作为对照组。实时荧光定量聚合酶链式反应检测(real-time polymerase chain reaction,RTPCR)胎盘组织中miR-30d 表达,免疫组化学染色检测胎盘组织中CHOP、激活转录因子 6(activating
transcription factor 6,ATF6)、葡萄糖调节蛋白78(glucose regulatory protein 78,GRP78)蛋白表达,
并分析miR-30d 与胎盘组织中CCAAT 增强子结合蛋白同源蛋白(CCAAT enhancer binding protein
homologous protein,CHOP)、ATF6、GRP78 等蛋白表达的相关性。采用t 检验、Pearson 相关系数分析
法进行统计分析。 结果  子痫前期患者胎盘组织中miR-30d 表达的2- △△ Ct 值为0.308±0.072,显著高
于对照组的0.115±0.014,差异有统计学意义(P<0.01)。子痫前期胎盘组织中CHOP、ATF6 和GRP78
ERS 标志蛋白表达较对照组显著升高,差异有统计学意义(P<0.01)。Pearson 相关分析显示子痫前期患
者胎盘组织中miR-30d 表达与CHOP(r=0.517,P<0.05)、ATF6(r=0.452,P<0.05)和GRP78(r=0.484,P<0.05) ERS 标志蛋白表达呈显著正相关(P<0.05)。 结论 子痫前期患者胎盘组织miR-30d 高表达,升高的miR-30d 激活了ERS,可能是导致子痫前期发生的原因。

Abstract: 【Abstract】 Objective To explore the relationship between micro-RNA-30d and endoplasmic reticulum
stress in placenta of patients with preeclampsia, and its clinical significance. Methods From October 2018 to September 2019, 30 cases of preeclampsia patients admitted to the Haikou Maternal and Child Health Hospital were selected as the research objects. Another 30 cases of normal delivery in our hospital were used as control. Real-time PCR was used to detect miR-30d. The expression of CCAAT enhancer binding protein homologous protein (CHOP), activating transcription factor 6 (ATF6), glucose regulatory protein 78 (GRP78) in placenta was detected by immunohistochemical analysis. The correlation between miR-30d and the expression of CHOP, ATF6 and GRP78 in placenta was analyzed. T test and Pearson correlation coefficient analysis were used for statistical analysis. Results The 2- △ △ Ct values of miR-30d in preeclampsia group was 0.308±0.072, which was significantly higher than that of 0.115±0.014 in control group with statistical significance (P<0.01). The expression of CHOP, ATF6, GRP78 in placenta of preeclampsia group was significantly higher than that of control group with statistical significance (P<0.01). Pearson correlation analysis showed that the expression of miR-30d in placenta of preeclampsia was positively correlated with CHOP (r=0.517, P<0.05), ATF6 (r=0.452,P<0.05) and GRP78 (r=0.484, P<0.05) expression in preeclampsia patients (P<0.05). Conclusion The high expression of miR-30d in placenta of preeclampsia patients activated endoplasmic reticulum stress, which may be the cause of preeclampsia
