Correlation between single nucleotide polymorphisms of ABCC1, SLCO2B1 genes andmontelukast sodium in the treatment of bronchial asthma in children of Han nationality

Abstract: 【Abstract】 Objective To evaluate the correlation between leukotriene receptor antagonist(LTRA) and single nucleotide polymorphism (SNP) of ATP-binding cassette sub-family C member 1 (ABCC1) genes and solute carrier organic anion transporter family member 2B1(SLCO2B1)genes in the treatment of bronchial asthma in children of Han nationality. Method A total of 100 cases with bronchial asthma of Han nationality aged 4-13 years who were treated in Xiamen Hospital of Traditional Chinese Medicine Affiliated toFujian University of Chinese Medicine from April 2019 to December 2021 were selected as the study objects. Thers119774 locus of ABCC1 gene and rs12422149 locus of SLCO2B1 gene of the cases were detected by generationsequencing, and the SNP distribution of the two sites was analyzed and compared. All enrolled cases weretreated with LTRA. Pulmonary ventilation function and pulse shock lung function were detected and asthmasymptom control level was evaluated before and after 12 weeks of treatment, and the correlation betweenSNP and efficacy was analyzed. Statistical methods were used by t test and χ2 test. Result Before and12 weeks after treatment, cases with CC asthma at rs119774 locus of ABCC1 gene were treated with montelukastsodium forced expiratory volume in the first second (FEV1) [(87.0±16.5) vs (93.2±15.1) L, t=-2.071,P=0.038], peak expiratory flow (PEF) [(77.1±16.9) vs (86.5±16.6) L/s, t=-3.233, P=0.001]and forced expiratory flow 25 (FEF25) of 25% vital capacity [(69.6±18.8) vs (78.1±19.9) L/s,t=-1.985, P=0.047] were significantly increased. The indicators of pulse shock were significantlyimproved (P<0.05). The pulmonary function index PEF of SLCO2B1 gene rs12422149 AA typeafter treatment was higher than that before treatment [(79.0±16.0) vs (93.0±9.4) L/s, t=-2.547,P=0.011]. The lung function indexes of GG type after treatment were significantly higher than those beforetreatment (P<0.05). Control rate before and after treatment with ABCC1 [59% (59/100) vs 78% (78/100),78% (78/100), χ2=20.964, P<0.001]; control rates of AA, AG and GG type at rs119774 of SLCO2B1 gene beforeand after treatment [53% (8/15) vs 80% (12/15), 93% (14/15), χ2=12.786, P=0.002] and [56% (23/41) vs 78%(32/41), 78% (32/41), χ2=6.583, P=0.037] and [73% (32/44) vs 77% (34/44), 73% (32/44)], χ2=6.471, P=0.039]were significantly improved. Conclusion There is a correlation between the effect of LTRA on the SNP ofABCC1 and SLCO2B1 gene in children with bronchial asthma of Han nationality.
