The effects of folic acid on OV90 cells global genome DNA methylation and cell behavioral

Abstract: Objective To investigate the effects of folicacid on the Global genome DNA methylation and cell behavioral of folic acid on ovarian cancer cell OV90. Providing theory basis for prevention and treatment of ovarian cancer.Method Ovarian cancer cells were cultured in vitro for three months and the changes of the Global genome DNA methylation, the cell growth rates, the cell cycles and the apoptosis were detected after treatment of OV90 cells with different concentration of folic acid. Ovarian cancer cells in high folic acid group were treated with 40 mg/L folic acid, cells with routine culture were served as control group, with the concentration of folic acid 4 mg/L and cells in the group were treated with no folic acid. Respectively, the methylation of genomic DNA was observed by MethyFlash Methylated DNA Quantification Kit(colorimetric), the cell grough rates were detected by CCK-8; Flow cytometry was employed to observe the changes of cell cycles and the apoptosis. Results The methylation of Global genomic DNA of OV90 increased with the concentration of folic acid; The cell grouth rates decreased obviously; Compared with control group, the cell percentage of G2/M phase in folic acid group decreased(P<0.05), the cell percentage of S phase increased (P<0.05), and there was no significant change in G0/G1 phase (P>0.05)and the cell apoptosis showed an increasing tendency. Conclusions Folic acid may increase the methylation of the Global genome DNA of ovarian cancer cell OV90 and inhibit the cell growth.
