摘要 【摘要】 目的 探讨体外成熟过程对后代睾丸组织母源印记基因差异表达和调节的影响。方法 本文建立单精子卵胞浆内注射术(institute for clinical systems impmvement，IcsI)和体外成熟(in vitm maturation，IvM)妊娠小鼠模型，利用实时定量PcR和焦磷酸测序技术检测后代睾丸组织中6个典型母 源印记基因的表达和甲基化状况。结果 IvM小鼠睾丸组织中母源印记基因的表达相对于IcsI小鼠存在 高表达和低甲基化现象。结论 体外成熟可以通过调节表观遗传学修饰影响睾丸组织中母源印记基因 的表达。

Abstract: 【Abstract】objectives The aim of this research was to study the effect of in vitro maturation on the expression and the regulation of matemal imprinted genes in the testis of male offspring．methods Institute for clinical systems impmvement(ICSI)and in vitro maturation(IVM)conceived mice were established，and Quantitative real time RT—PCR and Pyrosequencing were used to detect the expressions and methylation levels of 6 matemalimprinted genes in their testes．Results Compared with ICSI mice，the increased expressions and decreased methylation levels were shown in IVM mice．Conclusion IVM may innuence the expressions of matemal imprinted genes in testis by regulating epigenetic modification．
