摘要 细胞的发育是一个动态过程,染色质状态的变化、DNA表达等分子层面的调控支撑着细胞形态的变化和功能的完善。转录组测序技术(RNA-sequencing method,RNA-seq)是研究细胞发育的重要手段之一,能够获得细胞发育不同阶段全基因组的表达信息;但其结果只是细胞群基因表达量变化的均值,忽略了细胞的异质性。单细胞转录组测序技术(single-cell RNA-sequencing method,scRNA-seq)可以在单细胞水平获得全基因组范围的表达谱,有助于发现罕见细胞类型,并能更直观反映处于不同生长发育阶段的细胞中基因表达变化。虽然scRNA-seq已经取得了很多有意义的结果,但是该项技术操作还面临挑战;另外,scRNA-seq数据的处理与常规RNA-seq也有所区别,目前已经逐渐发展出针对scRNA-seq的计算机技术和算法。
