Abstract 【Abstract】 Objective To establish a cell model of mitochondrial disease in Leigh syndrome, the induced pluripotent stem cells (iPSCs) were obtained from the endometrium endometrial tissue of the patients with mtDNA 8993T>G mutation. Methods Endometrial tissue was obtained from patients cultured by primary culture method, transfected with Sendai virus in 3rd or 5th passages, which were transferred to feeder layer cells for further culture after the cells proliferated. On the 20th day, the clones were selected and iPSCs lines were established, and the multi-function and differentiation ability of the tumor were identified by alkaline phosphatase (AP) staining, immunofluorescence and teratoma experiments in mice. Results IPSCs were established successfully, AP staining was strongly positive after stable passage, OCT4,SOX2, SSEA-4 and TRA-1-60 were strongly expressed, teratoma experiments in mice showed that the iPSCs had the potential to differentiate into endoderm, mesoderm and ectoderm, and the iPSCs line was established successfully. Conclusions IPSCs are established in endometrial tissues of patients with mitochondrial disease Leigh syndrome by non-genetic integration method of Sendai virus. It provides a cellular model for the study of biochemical and molecular characteristics, drug screening and toxicology of Leigh syndrome pathogenesis
