摘要 【摘要】 目的 探讨脱蜡丝线在腹壁不同解剖层次内的线结诱导早期局部免疫应答的情况,并分析传统丝线蜜蜡涂层降解后的影响。 方法 采用10 ~ 12 周Balb/C 小鼠共18 只。4-0 带针慕斯线经脱蜡处理。每只小鼠的腹壁从左向右分3 组打结:皮下组、肌层组和腹膜外间隙组。术后1 周取材以线结为中心的全腹壁组织。免疫组织化学染色了解线结周围抗原肽- 主要组织相容性复合体(major histocompatibilitycomplex,MHC) Ⅰ / Ⅱ、CD4、巨噬细胞标志物F4/80、IL-6 的表达水平,测量积分光密度(integral opticaldensity,IOD)和面积(area)并计算出平均光密度(optical density,OD)。OD=IOD/Area。Masson 染色了解线结周围纤维化情况。统计学方法采用ANOVA 分析和student's t 检验。 结果 HE 染色可见线结准确位于腹壁不同解剖学层次和位置。免疫组织化学结果显示,线结周围抗原肽-MHC Ⅰ / Ⅱ类分子复合物的表达、CD4 分子表达、巨噬细胞浸润和IL-6 表达情况,皮下组表达最明显,腹膜外间隙组表达最轻微,3 组间差异具有统计学意义(P 值均<0.05)。Masson 染色显示,皮下组线结周围有较多胶原蛋白生成,而腹膜外间隙组线结周围胶原蛋白较少。 结论 传统丝线涂层降解后可诱导小鼠腹壁局部免疫应答,线结位于小鼠腹膜外间隙引起的炎症反应轻微。

Abstract: 【Abstract】 Objective To discuss early located immune response induced by knots of dewax-silk in different levels of mice abdominal wall, and to analyze the influence after wax degradation of conventional silk. Methods Eighteen mice (Balb/C, age 10-12 weeks) were used in this study. Silks (4-0 MERSILK?) were dewaxed. There were three knots in abdominal wall of the same mouse, which were respectivelysubcutaneous, intramuscular and extra peritoneal from left to right. After one week, the whole abdominal wall was sampled centering on knot. Immunohistochemistry staining was performed to detect the expression of
major histocompatibility complex (MHC) Ⅰ / Ⅱ , CD4, F4/80 and IL-6 around knots. Integral optical density (IOD) and area were measured to calculate average OD (OD=IOD/area). Masson staining was conducted to investigate the fibration around knots. ANOVA and student's t-test were used for statistical analysis. Results HE staining indicated that the knots were located accurately in subcutaneous, intramuscular and extra peritoneal cavity in abdominal wall. The immunohistochemistry staining indicated the expression ofMHC Ⅰ / Ⅱ , CD4+ T lymphocyte infiltration, macrophages infiltration and expression of IL-6, which werethe most obvious in subcutaneous group and the least obvious in extra peritoneal group (all P<0.05). The Masson staining showed that there was more collagen deposition in subcutaneous group than that in extra peritoneal group. Conclusions After the wax-coating degradation of conventional silks, located immune response is induced. If the knot is located in extra peritoneal cavity, the immune response is mild.
