Abstract: 【Abstract】 Objective To discuss early located immune response induced by knots of dewax-silk in different levels of mice abdominal wall, and to analyze the influence after wax degradation of conventional silk. Methods Eighteen mice (Balb/C, age 10-12 weeks) were used in this study. Silks (4-0 MERSILK?) were dewaxed. There were three knots in abdominal wall of the same mouse, which were respectivelysubcutaneous, intramuscular and extra peritoneal from left to right. After one week, the whole abdominal wall was sampled centering on knot. Immunohistochemistry staining was performed to detect the expression of major histocompatibility complex (MHC) Ⅰ / Ⅱ , CD4, F4/80 and IL-6 around knots. Integral optical density (IOD) and area were measured to calculate average OD (OD=IOD/area). Masson staining was conducted to investigate the fibration around knots. ANOVA and student's t-test were used for statistical analysis. Results HE staining indicated that the knots were located accurately in subcutaneous, intramuscular and extra peritoneal cavity in abdominal wall. The immunohistochemistry staining indicated the expression ofMHC Ⅰ / Ⅱ , CD4+ T lymphocyte infiltration, macrophages infiltration and expression of IL-6, which werethe most obvious in subcutaneous group and the least obvious in extra peritoneal group (all P<0.05). The Masson staining showed that there was more collagen deposition in subcutaneous group than that in extra peritoneal group. Conclusions After the wax-coating degradation of conventional silks, located immune response is induced. If the knot is located in extra peritoneal cavity, the immune response is mild.
