摘要   新生儿,尤其是早产儿,机体各组织器官功能尚未成熟,对外界环境的适应能力差,对疾病的抵抗力弱,感染后发病迅速,病死率高,因此,快速鉴定感染病原微生物对于新生儿重症监护病房中感染性疾病的防治尤其重要。目前常规病原微生物检测和鉴定主要为传统的分离培养和生理生化方法,这些鉴定方法周期长(3~5天),灵敏度和特异性较差,很难满足快速检测与鉴定的需求。虽然近年来使用免疫分析和分子生物学技术,能够实现病原微生物的快速、特异的检测和鉴定,但这些技术对检测人员自身素质要求高,操作复杂,并且无法进行后续药敏鉴定试验,往往只能作为常规病原微生物检测和鉴定的补充。随着蛋白质组学的发展,尤其是基于微生物表面蛋白分子量检测的基质辅助激光解析电离飞行时间质谱(matrix-assisted laser desorption/ionization-time of flight massspectrometry, MALDI-TOF MS)技术的日益成熟和广泛应用,MALDI-TOF MS已经逐步成为一种新型的微生物检测和鉴定技术,并且以其快速、准确、灵敏、高效、低成本、易操作等优势得到快速发展。本文将阐述MALDI-TOF MS分析技术在病原微生物检测和鉴定中的应用和前景。
