摘要 【摘要】 目的 研究SET/TAF-I 基因调控造血系统基因转录的机制,揭示SET/TAF-I 基因异常导致疾病的原因。方法 应用RT-PCR、Western blot 和原位杂交技术,检测造血细胞、白血病细胞和 斑马鱼胚胎中SET/TAF-I 基因的表达水平;应用免疫共沉淀技术,检测SET/TAF-I 与C/EBPα 的相 互作用。结果 SET/TAF-I 基因在骨髓增生异常综合征(MDS)细胞、急性髓系细胞白血病(AML) 细胞和淋巴细胞中,以及斑马鱼胚胎尾部造血组织中都有富集表达;SET/TAF-I 和C/EBPα 间存在相 互作用。结论 C/EBPα 是SET/TAF-I 靶基因,SET/TAF-I 通过C/EBPα 调控下游造血系统基因的转录。

Abstract: 【Abstract】 Objective To investigate the regulatory role of SET/TAF-I in normal and malignant hematopoiesis. Method The expression level of SET/TAF-I was respectively detected in MDS cells, AML cells, lymphocytes and zebrafish embryos by RT-PCR, Western blot and in situ analyses. The interaction between SET/TAF-I and C/EBPα was validated by Co-Immunoprecipitation (Co-IP). Result SET/TAF-I mRNA expresses in MDS cells, AML cells , lymphocytes as well as caudal hematopoietic tissue (CHT) at 3dpf in zebrafish embryos. SET/TAF-I binds to C/EBPα directly as indicated by Co-IP. C onclusion SET/TAF-I
might regulate the transcription of hematopoietic genes by interacting with C/EBPα.
