Abstract: 【Abstract】 Objective To explore the relationship between intestinal flora and intussusception based on Mendelian randomization (MR). Method Using summary statistics from Genome-wide Association Studies(GWAS) conducted by the MiBioGen and FinnGen consortia, a two-sample MR analysis was performed. Thecausal relationship between intestinal flora and intussusception was analyzed using methods such as inversevariance weighted (IVW), MR-Egger regression, weighted median estimator (WME), weighted mode, and MRPRESSO regression. Result A total of 3 631 instrumental variables from 211 microflora were included.A total of 5 types of intestinal bacteria were associated with intussusception detected by IVW method,among which Bifidobacterium had a reverse correlation with intussusception (OR=0.376, 95% CI: 0.157-0.899), Ruminococcus (OR=1.461, 95% CI: 1.058-2.807), Adlercreutzia (OR=1.893, 95% CI: 0.318-3.502),Allisonella (OR=1.715, 95% CI: 1.009-2.915), Clostridium (OR=1.974, 95% CI: 1.046-4.451) were positively correlated with the incidence of intussusception. The results of MR-PRESSO and MR-Egger showed that there was no heterogeneity and horizontal pleiotropy. Conclusion Bifidobacterium may reduce the risk of intussusception and postoperative recurrent intussusception. Ruminal cocci, Adlercreutzia, Allisonella, and Clostridium may increase the risk of intussusception
