摘要 Ⅰ 型 磷 脂 酰 肌 醇 4- 磷 酸 5- 激 酶（type Ⅰ phosphatidylinositol 4-phosphate 5-kinase，PIP5KIs）是体 内合成 4,5- 二磷酸磷脂酰肌醇［phosphatidylinositol 4,5-bisphosphate，PI(4,5)P2］的 关 键 酶，PIP5KIs 催化磷酸化 4- 磷酸磷脂酰肌醇（PI4P）肌醇环上第 5 位 产 生 PI(4,5)P2［1］ 。 已 有 研 究 发 现 PI(4,5)P2 是神经系统发育的重要调节因子［2-4］ ，作为脂质第 二信使的前体，PI(4,5)P2 被磷脂酶 C 水解产生两种 脂质第二信使：二酰基甘油（DG）和 1,4,5- 三磷酸肌 醇（IP3） ［5］ ，DG 激活蛋白激酶 C，IP3 通过 IP3 受体 使内质网释放 Ca2+ 而增加细胞内 Ca2+ 浓度。此外， PI(4,5)P2 被磷脂酰肌醇 3- 激酶（phosphatidylinositol 3-kinase，PI3K）磷酸化产生脂质第二信使 3,4,5- 三 磷 酸 磷 脂 酰 肌 醇［PI(3,4,5)P3］而 发 挥 其 调 控 作 用。PI(4,5)P2 通过上述第二信使进一步调节细胞 功能，参与肌动蛋白细胞骨架重组、膜运输、细胞黏 附、离子转运蛋白及通道的活性和凋亡等过程，参 与细胞生长、存活和凋亡［6］ ，从而调节神经系统的 发育。PI(4,5)P2 局部浓度的高低，对不同的细胞通 路及细胞功能至关重要［7-8］ ，而且每种 PIP5KI 调控 细胞内不同区域 PI(4,5)P2 池中 PI(4,5)P2 的含量。 PIP5KIs 通过调控 PI(4,5)P2 在细胞中的浓度，进而在 胚胎神经发育过程中的神经突起生长、神经元分化 及神经细胞迁移等方面发挥重要作用。本文将阐述 PIP5KIs 在神经系统发育性疾病中的作用及机制。
