转化生长因子-β/ 白血病抑制因子信号通路与儿童肝细粒棘球蚴成囊调控的相关性

摘要 【摘要】 目的  探讨转化生长因子-β(transforming growth factor-β,TGF-β)/ 白血病抑制因子(leukemiainhibitory factor,LIF)信号通路与儿童肝细粒棘球蚴成囊调控的相关性。 方法 2020 年1 月至2021 年6 月,就诊于新疆维吾尔自治区儿童医院普通外科的肝细粒棘球蚴病患儿中,选取40 例为研
究对象。行囊肿摘除术,体外分离原头蚴并培养。分为3 个研究组(分别添加LIF 因子、LIF siRNA、
Smad4 siRNA)及1 个对照组。培养2 周后,镜检具有成囊特征的原头蚴,提取总蛋白,采用酶联免疫吸
附试验检测TGF-β、Smad2、Smad3、Smad4 和LIF 的表达水平。统计学方法采用单因素方差分析和t
检验。 结果  各组间TGF-β、Smad2、Smad3、Smad4、LIF 的表达水平比较,差异均有统计学意义
(P 值均<0.001)。对照组上述5 个指标的表达水平分别为(3 152±276)ng/L 和(17.4±3.0)、(15.9±0.9)、(18.6±1.8)、(1 334 ±236)μg/L。LIF 组TGF-β、Smad2、Smad4、LIF 的表达水平[(4 100±173)ng/L 和(34.3±3.0)、(26.3±2.2)、(1 984±237)μg/L]均高于对照组(P 值均<0.05)。LIF siRNA 组上述5 个指标的表达水平[(2 151±136)ng/L 和(9.2±1.2)、(12.2±0.8)、(12.8±0.4)、(464±244)μg/L]均低于对照组(P值均<0.05)。Smad4 siRNA 组上述5 个指标的表达水平[(2 529±267)ng/L 和(5.7±1.3)、(10.1±0.7)、(9.8±2.0)、(246±53)μg/L]也均低于对照组(P 值均<0.05)。 结论 体外实验表明,TGF-β/LIF 信号通路可能与儿童肝细粒棘球蚴病原头蚴成囊调控具有相关性,原头蚴往成囊方向发育可能受LIF 调控。

Abstract: 【Abstract】 Objective To explore the correlation between transforming growth factor-β(TGF-β)/
leukemia inhibitory factor (LIF) signaling pathway and the regulation of cyst formation of hepatic
echinococcus granulosus in children. Method From January 2020 to June 2021, 40 children with hepatic echinococcosis were selected from the Department of General Surgery, Children's Hospital of Xinjiang Uygur Autonomous Region. Cyst removal surgeries were performed, and the protoscolices were isolated and cultured in vitro. There were three study groups which were added with LIF factor, LIF siRNA and Smad4 siRNA respectively, and one control group. After two weeks of cultivation, the protoscolices withcystic characteristics were examined under the microscope, and total protein was extracted. Enzyme-linked immunosorbent assay was used to test the expression levels of TGF-β, Smad2, Smad3, Smad4 and LIF.One-way analysis of variance and t-test were used as statistical methods. Result There were statistically significant differences in the expression levels of TGF-β, Smad2, Smad3, Smad4, and LIF among the groups (all P<0.001). The expression levels of the above five indicators in the control group were (3 152±276) ng/L and(17.4±3.0), (15.9±0.9), (18.6±1.8), and (1 334±236) μg/L. The expression levels of TGF-β, Smad2,Smad4, and LIF [(4 100±173) ng/L and (34.3±3.0), (26.3±2.2), and (1 984±237)] μg/L] in the LIF group were all higher than those in the control group (all P<0.05). The expression levels of the above five indicators in the LIF siRNA group [(2 151±136) ng/L and (9.2±1.2), (12.2±0.8), (12.8±0.4),and (464±244)] μg/L] were lower than those in the control group (all P<0.05). The expression levelsof the above five indicators in the Smad4 siRNA group [(2 529±267) ng/L and (5.7±1.3), (10.1±0.7),
(9.8±2.0), and (246±53)] μg/L] was also lower than those in the control group (all P<0.05). Conclusion Invitro experiments have shown that TGF-β/LIF signaling pathway may be related to the regulation of cyst formation of hepatic echinococcus granulosus in children. The development of protoscolices towards cyst formation may be regulated by LIF.
