摘要 摘要目的探讨母鼠孕前营养过剩对子代小鼠器官发育影响。方法将20只昆明种雌鼠随机分为对照组和实验组,分别给予标准饲料和高脂高糖饲料,连续喂养4周,制成母鼠孕前营养过剩模型;4周后雌、雄鼠合笼,受孕,孕期继续上述饮食;产后母乳喂养,3周断乳,哺乳期问母鼠仍按各自组别进食;子鼠断乳后均以标准饲料喂养,监测子鼠生后体格发育(体重和身长),测量子鼠成年早期(生后6周)心、肝、胰、脾、肾和脑脏器系数,并对各脏器进行病理形态检查。结果(1)经过4周单纯喂养,实验组母鼠体重、血糖显著高于对照组(P<0.05);实验组子鼠新生期(生后3天)及成年早期(生后6周)体重显著高于对照组(P<0.05)。(2)实验组子鼠心、肝脏器系数显著高于对照组(P<0.05),胰腺脏器系数显著低于对照组(P<0.05),脾、肾和脑组织脏器系数与对照组差异无显著性(P>0.05)。(3)HE染色,病理形态观察,与对照组子鼠比较,实验组子鼠心脏房、室壁厚度增加,横行心肌纤维截面积增大,纵行心肌纤维厚度增宽;肝细胞呈轻度水肿;脾脏红、白髓以及边缘区结构不清,白髓发育不良。结论母亲孕前营养过剩造成子代出生及成年早期肥胖,
并对某些器官造成形态学影响,后者可能是母亲营养过剩影响子代生后远期健康的解剖学基础,具体
机制值得深入研究。

Abstract: Abstract Objective To investigate the effects of prepregnancy overnutrition on organ development
of offspring mice. Methods A total of 20 female Kunming mice were randomly divided into two groups,
control and study group, which were fed with lab chow and high fat and high sugar diet, respectively. After feeding 4 weeks, all female mice were mated with male mice. After delivery, all mothers reared their pups freely. During pregnancy and lactation, mothers of the two groups were continuously fed their corresponding diet. After weaning, all pups were fed with lab chow. Physical development of offspring was monitored from birth to postnatal 6 weeks. Organ coefficients of offspring major organs were determined and the morphology of them was observed through HE staining under microscope at postnatal 6 weeks. Results 1After feeding 4 weeks, the average maternal bodyweight and blood sugar of study group were significantly higher than those of control groupP0.05 . During neonatal and early adulthood period, the body weight of offspring of study group was significantly higher than that of control groupP0.05.2The organ coefficients of heart and liver were both significantly higher in offspring of study group than those of control groupP0.05. Pancreas/bodyweight in offspring of study group was significantly lower than that of control groupP0.05 .There was no significant difference in organ coefficients of spleen, kidney and brainP>0.05.3Under microscope,compared with offspring of control group, in offspring of study group, thickness of atrial and ventricular wall was thicker, liver cells presented slight edema, and boundary of white pulp??red pulp and marginal zone in spleen was obscure Conclusions: Maternal overnutrition during prepregnancy results in high birth weight and obesity during young adulthood, and morphological changes of some organs in offpsring. These morphological abnormities might be the anatomical basis of influence of maternal prepregnancy overnutrition on offspring long-life health. The profound mechanisms are worthy of further study.
